Project summary:

The project is aiming to develop treatment methods for a difficult and challenging clinical problem connected to degenerative lesions of the articular joints by using multidisciplinary approaches such as tissue engineering (TE) and regenerative medicine to produce transplantable new organs for repair of osteochondral (OC) lesions. To achieve this, the project will employ the latest developments in materials science and nanotechnology, combined with state-of-the-art cell biology and imaging techniques to develop biomimetic osteochondral (OC) TE product for treatment of patients with degenerative lesions of joints. We hypothesize that the combination of 3D cell printing and 3D scaffold fabrication methods and the use of advanced biomaterials, somatic cells, and differentiation factors will allow us to produce biomimetic TE construct consisting of different zones corresponding to the cartilage, calcified cartilage and bony layers of the native organ. The proper somatic cells (MSC) as well as optimal synthetic and natural biomaterials will be selected to engineer simultaneously the cartilage and bone tissues in osteochondral defect. An advanced numerical modeling, and characterization methods will be also applied in development of a novel 3d zonal osteochondral construct with biomimetic structure, controllable porosity and suitable mechanical properties. In vitro and in vivo tests will be performed to assess the developed TE construct.
